It is a decoratively simple neoclassical two-storey building, with initially ground-floor side wings that later acquired an additional floor. The framed in marble rectangular openings are symmetrical. At the edge of the tiled roof, an elaborate parapet emerges, which bears the name of the hospital. The wings behind the original neoclassical building were constructed later.
It was built with a bequest from the professor of surgery and rector of the University of Athens, Theodoros Aretaeos, who donated his entire fortune. Aretaeos did not want a luxurious hospital, but a building that was durable, safe and with separated rooms for better hygiene. It was designed by Elias Angelopoulos and Ioannis Ioannou. The innovation in Greece at that time was that a surgical amphitheatre and outpatient clinic within the administration wing were designed from the beginning. After the Asia Minor Catastrophe, the Aretaeio Hospital provided medical care and soup kitchen for refugees.
